fear
After seeing last night's episode, I think that "Locke" (a.k.a. Smoke Monster) must feed off of fear. In other words, I think he must be able to control and manipulate others from fear. Jacob never seemed afraid of him, which I think was part of Jacob's power (and why the Smoke Monster couldn't kill him).
Anyhow, last night it seemed like "Locke" perceived Desmond as an enemy because Desmond was not afraid. And mems how the actual Locke was not afraid of the Smoke Monster either? He looked "into the heart" of the Smoke Monster, and the monster didn't kill him.
It was interesting to see the contrast between Desmond's calm, fearless attitude and Hurley's jittery "Dude, we don't want anyone hurt" conversation with "Locke."
